Each of the printing units weighs 5.5 tonnes, meaning the company has produced more than half a billion tonnes of the units since the launch of the 102 format in 1975.
The German manufacturer said the 102 series was its most successful product, shoring up a 75% market share for long-perfecting presses and 50% for straight models.
The 100,000th unit will form part of a Speedmaster SM 102 perfecting press destined for the Danagards Grafiska printshop in Sweden.
